To describe the NINC Olympus FV1000 confocal microscope in your paper’s methods section please modify this example: “Images were collected on an Olympus FV1000 confocal microscope. Fluorophores were excited with <list wavelengths, commonly used are 405nm, 488nm, 543nm, 633nm> and emission was collected in these <list wavelength ranges> wavelength ranges with a <magnification, commonly used are 10, 20, 40, and/or 60X> objective lens.”
Wavelengths of excitation and wavelength ranges for emission are in the metadata of the .oib files from the Olympus scope. Please contact Jeffrey LeDue (email@example.com) if you need help finding this information and tailoring the above statement.
The NINC Olympus FV1000 confocal microscope saves images in Olympus’ .oib format. Exporting tiffs and jpegs from the FV acquisition software is not recommended as this can cause loss of metadata (e.g. the physical scale of the image). Please download and install the Fiji versions of imagej. Fiji uses the Bio-Formats importer to work directly with .oib files.
